About Cagrilintide
Cagrilintide is a long-acting synthetic analogue of amylin (islet amyloid polypeptide, IAPP), the pancreatic peptide co-secreted with insulin by beta cells. Acylation extends its circulating half-life well beyond that of native amylin, which is why it is used in research designs that call for sustained rather than pulsatile receptor occupancy.
Its targets are the amylin receptor family — AMY₁, AMY₂ and AMY₃ — each formed by the calcitonin receptor (CTR) in complex with a receptor activity-modifying protein (RAMP1, RAMP2 or RAMP3). Because the shared core is the calcitonin receptor, research using cagrilintide frequently characterises activity at both amylin and calcitonin receptors, along with downstream cAMP signaling and the gastric-motility and satiety-signaling pathways reported in the preclinical literature.
Research Applications
Cagrilintide is used in research investigating amylin receptor pharmacology, RAMP-dependent receptor selectivity, calcitonin receptor cross-activity, and the kinetics of long-acting versus short-acting receptor agonism. It also appears in comparative work alongside incretin receptor agonists, where the question under study is how distinct receptor systems contribute to metabolic signaling in the same model.
